ABSTRACT

Objective To explore the influence of TAC protocol (pirarubicin,cyclophosphamide and docetaxel) and AC-T protocol (pirarubicin,cyclophosphamide and sequential docetaxel) on T lymphocytes cell subsets (included CD3+ T cells,CD3+ CD4+ T cells,CD3+ CD8+ T cells,CD3+ CD4+/CD3 + CD8+),natural killer (NK) cells and part of the quality of life in the peripheral blood of patients.Methods A total of 66 patients with breast cancer included 31 cases for TAC protocol (6 cycles of chemotherapy) and 35 cases for AC-T protocol(8 cycles of chemotherapy).Collected respectively before and after the last chemotherapy of peripheral blood,used flow cytometry technique to detect the proportion of T cell subsets and NK cells in peripheral blood,and compared the change of the part of the quality of life before and after chemotherapy.Results After chemotherapy of total of 66 patients CD3+ T cells,CD3+ CD4+T cells and NK cells of peripheral blood ((68.58± 11.03) %,(33.53 ± 8.84) %,(18.27 ± 10.65) %) significantly lower than before chemotherapy ((74.03±13.04)%,(38.42±9.79)%,(19.83± 10.19)%),the differences were statistically significant(t =4.296,3.387,4.092,P< 0.05).Grouping was TAC protocol with AC-T,T cell subsets and NK cells before chemotherapy differences had no statistical significance.But after chemotherapy,CD3+ T cells,CD3+ CD4+T cells,CD3+CD8+ T cells and Karnofky score((64.63±10.01)%,(31.19±7.41)%,(24.66±7.40)%,(68.63± 5.100)%) of peripheral blood on TAC protocol were more decreased than AC-T protocol((72.29± 10.78)%,(35.74± 9.60) %,(30.15 ± 11.12) %,(77.29 ± 4.58) %),the differences were statistically significant (t =2.99,2.15,2.35,2.237,P<0.05).CD3+ CD4+/CD3+ CD8+ and NK cells were also decreased,but without statistical significance.The body quality of 66 patients was slightly increased after chemotherapy,but without statistical significance.Conclusion The body's immune function and the part of quality of the breast cancer patient's life have declined by chemotherapy drugs in varying degrees.Compared TAC,AC-T protocol has less influence on the immunological function and the part of living quality of patients with breast cancer.

ABSTRACT

Objective:To investigate the correlation between ultrasonic elasticity score ,elasticity coefficient and autoimmune thyroid function in patients with HT ( hashimoto thyroiditis ) .Methods: Used GE LOGIQ3 type Color Doppler Ultrasound Diagnostic System to acquire satisfactory elastosonography from 216 patients with HT,and evaluated the elasticity score and elasticity coefficient;to detecte the thyroid function and the titer of Thyroid peroxidase antibody ( TPOAb ) , Thyroid globulin antibody ( TgAb ) by chemiluminescence ,respectively investigated the correlation between ultrasonic elasticity score , elasticity coefficient and autoimmune thyroid function.Results:Serum Thyroid stimulating hormone (TSH),TGA and TPO in 216 patients with HT increased significantly , TgAb and elastic coefficient were positively correlated , r=0.54 , P<0.01;TgAb and elasticity score were positively correlated , r=0.496 ,P<0.01;TPOAb and elastic coefficient were positively correlated ,r=0.77 ,P<0.01 ,TPOAb and elasticity score were positively correlated,r=0.73,P<0.01.Conclusion: Real-time ultrasound elastography in combination with measurement of autoimmune thyroid function was helpful to diagnosis of HT .

ABSTRACT

Background and purpose:EPHA2 has been reported to enhance the proliferation of gastric cancer cells through promoting CyclinD1 expression and cell cycle progression. However, the underlying mechanism that EPHA2 promotes CyclinD1 expression and cell cycle progression is still poorly understood. Akt/mTOR signaling pathway has been reported to enhance the proliferation of gastric cancer cells by promoting CyclinD1 expression and cell cycle progression, and some studies have shown that EPHA2 can activate Akt/mTOR signaling pathway. Based on this, this study investigated whether EPHA2 promoted gastric cancer SGC-7901 cell proliferation through enhancing Akt/mTOR signaling pathway.Methods:Western blot was used to determine the effect of EPHA2 overexpression or knockdown on the phosphorylation of Akt and mTOR in SGC-7901 cells. SGC-7901-NC infected with control lentivi-rus and SGC-7901-EPHA2 cells with EPHA2 overexpression were treated with DMSO, MK2206 (an Akt inhibitor) and RAD001 (a mTOR inhibitor) for different time periods, respectively. Cell proliferation was detected using the CCK8 assay. Cell cycle was detected using lfow cytometry, and the expression of CyclinD1 was determined by Western blot. Results:Overexpression of EPHA2 enhanced Akt/mTOR signaling pathway in SGC-7901 cells, and silencing EPHA2 in SGC-7901 cells inhibited Akt/mTOR signaling pathway. MK2206 and RAD001 antagonized the promoting effect of EPHA2 on the proliferation, S-phase and CyclinD1 expression of SGC-7901 cells, respectively.Conclusion:EPHA2 promotes SGC-7901 cell proliferation through enhancing Akt/mTOR signaling pathway. Akt inhibitor or mTOR inhibi-tor could be an effective treatment strategy for gastric cancer patients overexpressing EPHA2.

ABSTRACT

Objective To establish the quality control standard for Yi-Li-Kang tablets. Methods Astragalus membranaceus and Herba Epimedh were identified by TLC. The content of Icariin was determined by HPLC. Results The linear range of Icariin was 0.288~2.016 μg. The average recovery was 100.83%, and RSD was 1.82 (n=5). Conclusion The method is simple, accurate and reliable, which can be used to control the quality of Yi-Li-Kang tablets.

ABSTRACT

OBJECTIVE:To explore the effect of Shugan jianpi formula on the proliferation and apoptosis of the hyperplastic tissues of mammary gland of model rats.METHODS:The rats were injected with benzestrofol and progesterone to establish mammoplasia model,meanwhile which were treated intragastrically with Shugan jianpi formula for 30 days.The degree of mammoplasia,as well as the PCNA(proliferating cell nuclear antigen),the apoptotic related regulating factors such as Bcl-2 and Bax and the apoptotic index(AI)of the mammary gland were detected.RESULTS:Shugan jianpi formula inhibited the mammoplasia,down-regulated the expression levels of PCNA and Bcl-2,up-regulated the expression level of Bax and AI,inhibited the proliferation of the mammary tissues and promoted apoptosis.CONCLUSION:Shugan jianpi formula blocks or reverses the hyperplasia of mammary gland by regulating the cell proliferation/apoptosis of the hyperplastic tissues of mammary gland.

ABSTRACT

Objective To investigate the prevention and treatment mechanism of liver-smoothing and spleen-srengthening formula on hyperplasia of mammary gland,we study the effects of it on morphology,histology,serum sex hormone and its receptor of rats with hyperplasia of mammary gland.Methods Sixty female Wistar rats(SPF)were randomized into 5 groups:control group,disease model group,the tamoxifen group,small dose of medicine group and large-dose drug group.Estradiol Benzoate and Progesterone were used to establish animal model of breast hyperplasia.Rats in different groups were respectively maintained with Tamoxifen,small and large dose of liver-smoothing and spleen-srengthening formula.Then changes of morphology,histology,serum sex hormone and its receptor were obtained.Results Liver-smoothing and spleen-srengthening formula could reduce the number of breast lobular and the expansion degree of adenovirus catheter.It also narrowed the acinar cavity.In serum of rats with breast hyperplasia,Estradiol(E2)and PRL were declined ;Progesterone(P)and Testosterone(T)were increased;FSH and LH were partly increased.Positive expression degree of ER and PR were inhibited obviously.Conclusion Liver-smoothing and spleen-srengthening formula can reduce not only the content of estrogen receptors(ER)and progesterone receptors(PR)but also their sensitivity to estrogen of rats with breast hyperplasia by regulating the serum sex hormone.Our studies suggest that it can obviously inhibit hyperplasia of mammary gland.

ABSTRACT

Objective To research the mechanism of treating cyclomastopathy with Runingshuang by measuring the blood rheology and mammary microcirculation. Methods 70 female Wistar rats of SPF were randomly divided into seven groups:the blank control group, the pathologic model group, the comparative group of tamoxifen ointment, the comparative group of Sanjierupi Glue, three remedial groups with large, middle and small dosage of Runingshuang. Pathologic model of mammary hyperplasia was made by injecting estrogen and progesterone. Except the former two groups, each group was treated with different drugs. The blood rheology and mammary microcirculation were measured after 30 days.
